Effect of Sinefungin and 5'-S-Isobutyl-5'-Deoxyadenosine on Immune Function of Spleen Cells from BALB/c Mice




Abstract
The effects of sinefungin and 5'-S-Isobutyl-5'-Deoxyadenosine(SIBA0 on in vitro mitogen induced proliferative response, cell cycle and antibody response has been studied in spleen cell cultures from BALB/c mouse.
Sinefungin and SIBA, inhibitors of methyltransferases inhibited the proliferative response to lipopolysaccharide(LPS) or concanavalin A(Con A) in a dose dependent manner. A time course study indicate that proliferative response was inhibited when
sinefungin was added within the first 12hr of culture, whereas the inhibitory effect of SIBA was showed in all groups tested. The fractions of S and G2+M phase of cell cycle were decreased by sinefungin and SIBA. In vitro polyclonal and T-cell
independent antibody responses were inhibited by sinefungin and SIBA. The inhibitory effect of SIBA on polyclonal response was stronger than that of sinefungin with the being dependent on the time of addition and exhibiting maximum effect at the
inhibition of cultures(0 hr). Sinefungin did not affect the polyclonal response when added after 36 hrs, but strongly inhibited LPS induced proliferative and polyclonal responses when lymphocyte was cultured with the inhibitors for more than 3
days. The
Ki values of for protein methylase I and III in LPS or Con A treated lymphocytes showed 3 to 100 times lower than those of SIBA.
It is demonstrated that exposure to both inhibitors of methyltransferases brought a marked suppression of some splenocyte functions at the concentrations having no effect on viability.
